Project title: COMBINING CHEMOTHERAPEUTIC EFFECTS OF FLAVAN-3-OLS WITH RNA INTERFERENCE TARGET THERAPY IN CANCER


Project summary:

Flavan-3-ols were studied in recent years mostly for their ability to reduce oxidative stress and less in terms of cellular and molecular mechanisms of action or the relationship between chemical structure and antitumor properties. The first objective of the project is to identify the compound with maximum efficiency and determine the optimal dose of therapy with minimal side effects. Another objective of the project follows the assessment of direct and indirect action on genes involved in modulating apoptosis post-treatment with flavan-3-ols, in the presence, respectively in the absence of p53 siRNA. We will continue with the identification of signalling pathways that interfere with apoptosis and reversing processes of blocking apoptosis in tumor cells, especially the effect of co-activation of apoptotic processes by flavan-3-ols in the presence of p53 siARN.

The objectives of this project are:
1. Toxicity studies to determine the optimal dose for therapy
2. Functional genomics studies for revealing genes involved in modulating apoptosis by flavan-3-ols in presence, respectively in absence of p53 siRNA.
3. Genomic studies to validate genes of interest. Correlation of gene expression data with those obtained from the validation at protein level. Highlight new therapeutic implications in cancer therapy by combining the chemotherapeutic effect of flavan-3-olis with RNA interference targeted therapy.
